چکیده

The t(12;21)(p13;q22) results in the fusion of the TEL gene located on chromosome 12 with the AMLI gene located on the derivative chromosome 21. Because this translocation is difficult to detect using standard cytogenetic techniques, 27 previously karyotyped Blineage acute lymphoblastic leukemia (ALL) cell lines were evaluated for the presence of the EL-AMLI fusion using the reverse transcriptase-polymerase chain reaction (RT-ER), fluorescence in situ hybridization (FISH), and cDNA sequencing. Six cell lines expressed the TELAML1 chimeric transcript by RT-PCR and the t(12;21) was confirmed by FISH analysis with probes for TEL, AMLI, and chromosome 12. While only one of the 6 cell lines with the t(12;21) lost the der(l2)t(l2;21)-encoded AMtl-TEL fusion transcript, 4 cell lines lacked expression of the non-

Breakage and fusion of the TEL (ETV6) gene in immature B lymphocytes induced by apoptogenic signals.

TEL-AML1 fusion resulting from the t(12;21)(p13;q22) is one of the most common genetic abnormalities in childhood acute lymphoblastic leukemia. Recent findings that site-specific cleavage of the MLL gene can be induced by chemotherapeutic agents such as topoisomerase-II inhibitors suggest that apoptogenic agents can cause chromosomal translocations in hematopoietic cells. This study demonstrate...

Gene expression patterns associated with recurrent chromosomal translocations in acute lymphoblastic leukemia.

We obtained a global view of gene expression in both cell lines and pediatric acute lymphoblastic leukemia (ALL) samples that harbor one of several selected chromosomal abnormalities. When the cell lines were studied alone, we found that these chromosomal abnormalities were associated with the predominant variation in transcriptional programs across the set of cell lines studied. When cell line...

Small ubiquitin-like modifier conjugation regulates nuclear export of TEL, a putative tumor suppressor.

Posttranslational modification by small ubiquitin-like modifier (SUMO) conjugation regulates the subnuclear localization of several proteins; however, SUMO modification has not been directly linked to nuclear export. The ETS (E-Twenty-Six) family member TEL (ETV6) is a transcriptional repressor that can inhibit Ras-dependent colony growth in soft agar and induce cellular aggregation of Ras-tran...

Structure and possible mechanisms of TEL-AML1 gene fusions in childhood acute lymphoblastic leukemia.

TEL-AML1 gene fusion derived by chromosomal translocation is a common acquired genetic lesion in pediatric cancer that is present in approximately 25% of B-cell precursor acute lymphoblastic leukemias, and recent evidence suggests that this recombination event may initiate leukemogenesis prenatally during fetal hemopoiesis. Analysis of the DNA sequence and structure surrounding the breakpoints ...
